Can I test another GPU in a free PCIE 16X slot without removing my main water cooled GPU in the first PCIE 16X slot? Will the water cooled GPU remain detected with no PSU power and DP connection?

Yep, it will.
Also you need to read your motherboard manual carefully. Mine (Maximus XI Extreme) can't get x16 for 2nd pcie slot at all, so if you get a single gpu and insert into slot 2 - it will work as x8, if slot 1 enabled at least (but not occupied at all).
But there is options to turn off slot1 and use its lines for DIMM M.2 module, maybe in that case I could get x16 on 2nd pcie slot, but I never tried that.

Depending on your motherboard some have the switches to disable a pcie slot. However that being said if your motherboard will boot with a gpu in the second slot and you remove the power to your top card it should work to my knowledge.

But that will work as x8, not as x16 if slot is enabled and gpu even without power connection occupied that slot.

According to his post he only wants to see if the gpu is working, not keep it in that slot full time. But we don't know what his Mobo and CPU even are to Know what speed the slot will run at.

oh, I think I misread his post.
I read x16 slot as "I want run my gpu at x16!", and that is not what he said :)
 
 So yeah, even if his gpu will turned on - he could work with 2nd gpu simply plugging his monitor to it.
I had similar testing with crossfire setup - by switching output gpu with a monitor connection I found which of my gpus are bad (artifacts).
Not sure did I just switched ports or rebooted too though, but thats not a big deal.

To only test only the secondary gpu simply disable the first gpu inside device manager once inside windows, and make sure your dp connection is only connected to the secondary card.

Could I plug in a pair of PCIE cables from a spare PSU to one of the GPU's?

Yeah, I'm curious about this one to. Seems like PS_ON for both PSUs have to be turned on at the same time? What's the tolerance for differences in the timing for both PSUs being powered on? Is it in seconds or milliseconds? The second card would probably be getting some weird supply as 75W comes through the motherboard slot and the remaining from the other PSU.

The other option would be if you have a friend or family who would let you test the card in their system but with the shelter in place orders it's probably best to take apart the tank and set it back up again.

A Phanteks P600S is very heavy with a full custom loop. It sits on the desk in the corner of the room so has to be taken down to gain access to the back with the wiring. I will manage, have the time in lock down. Would it be recommended to remove the second pair of PCIE cables once finished with them or perhaps leave them there for future use?

I've got the second gpu plugged in but I don't have full screen resolution and only 60hz. I'm running PX1 oc scan, it's hitting power target but gpu clocks aren't rising. Is this because I'm in 8x? How do I get full resolution?

Hitting the power limit with the gpu clock stuck at a low clock isn't good. Sounds like the card has issues. It's not because you're at x8. I'd try ddu'ing the drivers.

Never mind, I got the card to clock up with 3D Mark firestrike, the oc scan was just taking ages to clock up. Card looks to be functioning normally...

The card with air cooler had no room to breath crammed into the lower PCIE slot underneath the GPU/waterblock still in the top PCIE slot. The card got up to 80C on the core during the OC scan so I Pulled the plug. I hope I didn't hurt it... I manged to keep it cooler in 3D Mark to assess it's function. Ran through several of benchmarks just fine.

Shouldn't have to worry about 80c or even 90c since it wil protect itself.  Normal use of operation in the 70c-80c+ range if you're on air.
