The GTX 480 will include a 2x6-pin PCIe to 1x8-pin PCIe adapter, so yes if you use that then you'd be fine. You'd use that one adapter which will use up 2x6-pin PCIe plugs, and you'd need a 3rd PCIe 6-pin to the GTX 480.
If you went SLI you'd have to use 2x6-pin to 1x8-pin twice, leaving you no more 6-pin PCIe connectors, but the cards will also include 2xmolex to 1x6-pin PCIe adapters. I wouldn't recommend that however, as PSUs that don't have 8-pin connectors generally aren't going to be sufficient to run newer high-draw GPUs like the GTX 480.
Also, don't try and use the EPS 8-pin for a graphics card, its keyed differently for a reason.
