The missing pin 20 of the 24-pin connector was for a -5V that's no longer used, not an issue there.
You should plug in the 24-pin connector and CPU power connectors..You don't need a CPU or RAM or anything installed, all you need is the PSU switched on to supply power to the mobo and then the Standby LED should be on telling you the mobo is getting power.
You need to format a USB flash drive to FAT32 and use the BIOS Renamer utility if you have an ASUS mobo..Everything you need to do should be explained in your mobo manual.
If you expect any specific/meaningful help, you need to say what all harware/mobo you have.
