I just got a evga geforce gtx 1080 sc gaming acx 3.0 to replace my old RX 480.  When I install it I hear the POST single beep but have no signal on my monitors.  These are known good cables I used for the 480.
Asus m5a78l-m-usb3 latest firmware
AMD FX 6300

    I see some Asus M5A78L-M/USB3 builds with new/newer NVIDIA cards at UserBenchmark, so compatibility shouldn't be an issue.

    Try turning off/unplugging the PSU and clearing CMOS..Turn off/unplug the PSU and reseat the card/all connectors fully.

    Try some of the other different video outputs on the card or try your card in another PC if possible..You could always take the card to your local PC repair shop for real quick testing..Maybe the card is no good.

    Did you use DDU to uninstall your old drivers before physical installing the newer GPU?

    I have no signal at POST so the old drivers aren't in play at that point.

    If I might ask how you acquired this GPU? Did you do a full reset on the BIOS? (remove battery hold CMOS for 30 seconds let set for 5 minutes) Is this a known good GPU that works in another system? Do you have another system to test it in? If acquired from a credible source, I'd be looking for a refund. If not, well good luck.

    Should remove old AMD drivers first ... install old GPU, run DDU safe mode & shut down, then install new GPU
     
    Is Windows Fast startup - Off ? - the last file had your "old GPU" info in it
     
    If the hardware is working --> Might find a solution in this
